Pathologic Study of Suramin Effects on the Cell Line of Penile Squamous Cell Carcinoma



Abstract
Suramin, a polyanionic compound with antiparasitic, activity, has been recently shown to have growth factor antagonist properties and to affect growth matastic lesions of several malignancies. Chung-Ang University Penile Cancer Cell

from
human penile squamous cell carcinoma has strong tumorigenicity when implanted into the nude mouse.
Herein, an experimental study was done to investigate the effect of suramin on the in vitro and in vivo proliferation and metastasis of CUPE-1, morphological changes of CUPE-1 cells induced by-suramin.
Suramin induced vacuolar degeneration and cell death as well as increased myelin figures and defects of desmosomes in CUPE-1 cells on light and electron microscopy, respectively. EGF immunostaining revealed positive intracytoplasmic domain both
in
control and suramin treated groups. Weekly intraperitioneal adminstration of 40gm of suramin to nude mouse inhibited the proliferation and metastasis of intraperitioneally implanted CUPE-1 cells, significantly(p<0.001), but did not show
significant
effect on the proliferation of subcutaneously implanted CUPE-1 cells.
There results suggest that suramin inhibits the growth and metastasis of the cell line of penile squamous cell carcinoma, and the main pathologic effects of suramin against CUPE-1 is manifested as vacuolar degeneration, cytoplasmic myelin figures
and
defects in desmosomal structures.
